When I do order, it's usually because I need one or two specific parts. I usually search for stores which have those parts in the quantities I need, at the best price.
I usually have parts/figures/sets on my wanted list which I watch to see if they ever come up for a good price. I then will filter those on the store, and see if there's anything else I've been looking for that the store has.
Finally, I will poke around the store a bit. Often I'll set the filter to lowest price to see if there's any good deals. I generally go through the Grey and Dark Grey categories to see if there's anything good.
Bricklink becomes easier to navigate as you use it more. Now I can find whatever I want fairly quickly, but at the beginning it was tedious and I spent time in chat having a few friends (who are sellers) help me find things.
If you want to spend some time at it, there is a method to find stores with all the parts you want. You can create a coded file with the item number, colour number, quantity, etc., of each part you want, and it will automatically select the "best match" stores for you. I tried it once, and I found the process of finding all the number codes too lengthy for it to be worth it.
...speaking of that, I need to buy some blue bricks. Have fun.

I will use it when I get a "real job".
PS: Talking with CSF guys at IRC, I calculated how much money I would need inorder to visit this years BrickFest. Only hefty 1300 € which is 1 572 dollars.)

Bricklink Is great! Everything has to be bought straight away which Is good, Its not like eBay, There are no auctions so Its easier and better to buy stuff from there.
